Get the odor: Characterization of improved CAR variants
More than probably any other sense, smell evokes memories and aldehydes are the key ingredients for creating these moments. Bioreduction of carboxylic acids to aldehydes by Carboxylic Acid Reductases (CARs) show promising results for a more sustainable route to aldehydes. CARs are, for example, industrially used for vanillin production. Just recently, structural data of these multi-domain enzymes was published. Our research group combines extensive knowledge for generating improved enzyme variants.
In this project, we are specifically interested in investigating the characteristics of these enzyme variants. Therefore, cloning and heterologous expression will be the starting point to allow enzyme purification and characterization. Investigation of the substrate scope in vivo and in vitro will enable to assess the potential of these new enzymes for biotechnological application.
The ideal master candidate studied molecular biology, biochemistry or biotechnology and has very good theoretical and practical knowledge of: molecular biology methods (PCR, DNA extraction and purification, gel electrophoresis, cloning); bioinformatic tools (BLAST, CLC-workbench); microbiological work such (bacteria cultivation on small scale, cell disruption, protein purification, NuPAGE analysis and enzyme activity assays with HPLC and GC analysis). However, knowledge of all techniques is not mandatory. Applicants should be enthusiastic to reach defined goals, to be motivated to learn new techniques and to present their results.
